Analysis Note

ControlIncluded free of charge with the antibody is 40 µg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 µL of sterile deionized water. For negative control, preincubate 1 µg of peptide with 10 µg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.

In preparing full-length protein samples for Western blot or immunoprecipitation, the procedures should be performed at 4°C, with the following protease inhibitor mixture: pepstatin A (1 µg/mL), leupeptin (1 µg/mL), aprotinin (1 µg/mL), Pefabloc SC (0.2 mM), benzamidine (0.1 mg/mL), and calpain inhibitors I and II (8 µg/mL each).

Immunoprecipitation

Immunocytochemistry. Recommend subsequent staining with a labeled second antibody.

Western blot: 1.5 to 3.0 µg/mL (1:100-1:200) using ECL

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.

Immunogen

Purified peptide CNB1 from 851-867 of alpha 1B subunit of rat brain voltage-gated calcium channel (Accession Q02294; Westenbroek et al. 1992) containing N-terminal lysine and tyrosine, conjugated to KLH with gluteraldehyde (Harlow & Lane 1988).

Physical form

Affinity purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 1% BSA and 0.05% sodium azide as a preservative. Reconstitute with 200 µL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Specificity

Recognizes a 1B subunit (both LMW and HMW forms) of voltage-gated calcium channel. Does not cross react with any other calcium channel antigens tested so far.

SPECIES REACTIVITIES: It is expected that the antibody will also work on mouse due to sequence homology (15/17). Other species have not been tested.

Storage and Stability

Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at 2-8°C in undiluted aliquots for up to 2 weeks. For longer periods, it is recommended to add glycerol (ACS grade or better) at a ratio of 1:1 and store the antibody at -20°C for up to 6 months. Avoid repeated freeze/thaw cycles.
